Thousands of inmates began rioting in Damanhur prison, breaking down cell doors and attempting to escape the detention facility 150 kilometres (93 miles) north of Cairo, the official said.
"Two prisoners were killed and at least ten were wounded by gunshots," he said.
Twelve people were killed in riots at the same prison on January 28 at the start of the revolt against president Hosni Mubarak which led to him standing down on February 11, the official said.
The latest riot came as the head of security of Beheira province, to which Damanhur belongs, was transfered after videos appeared on the Internet apparently showing him saying Egypt`s security forces were still "masters" of the country.
Egypt`s new military rulers, who took power after Mubarak resigned, have vowed to prosecute any official found guilty of abuse.
Former interior minister Habib al-Adly is to face trial on March 5 as part of a large probe into corruption launched after the uprising. (*)
